Balancing Act: Understanding the Interactions Between CBD and Ibuprofen
In vivo, DH-CBD potentiated IGly evoked by puff application of 30 uM glycine in spinal dorsal horn neurons of rats. Moreover, DH-CBD significantly suppressed inflammatory thermal and mechanical pain hypersensitivity induced by intraplantar administration of 20 ul CFA (1:4 in saline) into one paw in rats (Fig. 1 A). This analgesic effect of DH-CBD was significantly blocked by a glycine receptor antagonist and was not due to its agonist activity at CB1 or CB2 receptors.
DH-CBD specifically targeted the a3 GlyRs in spinal dorsal horn neurons and was effective in treating neuropathic pain. It is likely that a combination of a3 GlyRs and CB1 and CB2 receptors mediates the glycinergic cannabinoid-induced analgesia. This suggests that the a3 GlyRs could represent a new target for the treatment of chronic pain in neuropathic disorders such as peripheral neuropathy.
